Tolkien felt the events occurred a little over 6,000 years ago (Letter 211). I haven't found any evidence of an attempt by Tolkien to tie his imaginary history in with Biblical or other hitsories. However, Gondor and Arnor would have been well to the north of Sumer, and 1,000 years could easily mark the decline of both realms, though not their demise.
